Scientific background: Human Annexin A8 was originally identified from a 2-kb cDNA transcript containing an open reading frame that encoded a 327-amino acid protein termed vascular anticoagulant beta. It is a minor annexin in human placenta and shows restricted expression in lung endothelia, skin, liver, and kidney. The gene is selectively overexpressed in acute myelocytic leukemia. The Annexin A8 gene is mapped to chromosome 10q11.2 by fluorescence in situ hybridization. Affected human promyelocytes contain a nonrandom chromosomal translocation breakpoint involving chromosomes 15 and 17 and creating 2 hybrid mRNA fusion products and chimeric proteins. These involve the retinoic acid receptor-alpha geneon chromosome 17q12 and the promyelocytic leukemia gen on chromosome 15q22. Strong overexpression of Annexin A8 in this disorder could be repressed by retinoic acid-induced expression of the RARA gene product.
