Hongotoxin-1. A Potent Blocker of Some K_v1 K⁺�Channels.
Hongotoxin was originally isolated from the scorpion Centruroides limbatus and was shown to block cloned and heterologously expressed (in HEK 293 cells) K_v1.1, 1.2 and 1.3 with IC₅₀ of 31, 170 and 86 pM respectively. In addition, it blocks K_v1.6 with low affinity (IC₅₀ = 6nM). Hongotoxin-1 action was examined by monitoring radiolabeled Rb+ efflux from K_v channels-transfected HEK 293 cells. Hongotoxin-1 blocks 125I-Margtoxin (Margatoxin) binding to rat brain synaptosomes or 125I-Kaliotoxin binding to purified K_v1.3-KcsA chimeras. A mutated 125I-Hongotoxin-1 was used to immunoprecipitate (with specific K_v antibodies) rat brain K_v channels and to determine their subunit composition.
