IL-33 belongs to the IL-1 family and is closely related in structure to IL-18 and IL-1beta. IL-33, IL-1beta, and IL-18 are synthesized as biologically inactive precursors and are cleaved by the enzyme caspase-1 to be secreted as active mature forms. IL-33 stimulates target cells by binding to the IL-1R/TLR superfamily member ST2 and, subsequently, activates NF-kappaB and MAPK pathways via identical signalling events to those observed for IL-1beta. In addition, IL-33 is a nuclear factor (NF-HEV) abundantly expressed in high endothelial venules from lymphoid organs that associate with chromatin and exhibit transcriptional regulatory properties.

Dilutions: Each lot of this protein is quality control tested by ELISA assay. For use as an ELISA standard, a standard curve comprised of doubling dilutions from 1000 pg/ml to 15.6 pg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
Storage buffer: Upon receipt, store unopened vials at 4C immediately and use within 12 months from date of receipt. Prior to use, reconstitute the lyophilized powder with 0.2 ml of PBS containing a carrier protein (e.g., 1% BSA, protease free), pH7.4. Re-cap vial, vortex. Allow the reconstituted standard to sit at room temperature for 15 minutes, vortex again to mix completely. The reconstituted standard stock solution can be aliquoted into polypropylene vials and stored at -70C for up to one month. Do not re-use diluted standards. Use a manual defrost freezer and avoid repeated freeze thaw cycles.
