SPECIAL FEATURES: 1. Clear and sharp bands using a recombinant protein ladder ranging 14 to 100 kDa. 2. No need to use additional secondary antibodies or reagents in addition to those for detection of proteins of primary interest in Western blots. 3. Compatible with colorimetric, chemiluminescent, HRP and AP development for signal detection. 4. Premixed with sample loading buffer and preheated, ready for use. PROCEDURE FOR DETECTION OF IgY LADDER DURING WESTERN BLOTTING: 1. Load 10 microl of IgY ladder on the minigel (adjust volume for larger gels) adjacent to proteins of interest in separate lanes. 2. After SDS polyacrylamide gel electrophoresis, transfer the proteins to a PVDF blotting membrane. 3. Block membrane with 5% non-fat milk in TBS-T (Tris-buffered saline containing 0.05% Tween-20, pH 7.4) for 1 hour at room temperature, or longer at 4(o)C. 4. Incubate membrane with your IgY primary antibodies appropriately diluted with 5% milk in TBS-T @ R.T. for 1 h. 5. Wash 3 x 3-5min with TBS-T. 6. Incubate with secondary antibody (e.g., GAYFC--HRP) at dilution 1:1,000 for colorimetric assay or 1:10,000 - 1:100,000 for ECL (with 0.5% milk TBS-T) @ R.T. for 1 h. 7. Wash 3 x 3-5min with TBS-T 8. Perform color development, or perform ECL signal detection.
