Calcicludine. Neuronal L-type Cav Channel Blocker. Calcicludine is a natural peptide isolated from the Dendroaspis angusticeps green mamba venom, by a modification to the procedure of Schweitz et al and purified to homogeneity. Calcicludine was shown to be a specific neuronal L-type Cav channel blocker and was shown to inhibit other neuronal HVA Cav channels (N and P type) as well. 1 μM Calcicludine had no effect on skeletal muscle Cav channels (Cav 1.1) as well as on T-type (Cav 3) channels in heart and neurons. Calcicludine was tested to confirm its ability to block spontaneous or K+-induced contraction of cardiac cells. It blocks Cav 1.2 cloned channels with IC50 of 88 nM and had very little effect on Cav 2 family channels. In addition the block was irreversible upon wash and never complete but reached a maximum of 58%. Ion Channel Modulators. Voltage - Gated Ca2+ Channel Blockers.
