ELISA / Assay / CLIA Kits
One standardized format for detecting the presence of an antigen in a liquid sample is the enzyme-linked immunosorbent assay (ELISA). Commercially available ELISA kits provide the necessary calibration samples, antibodies that will react to the antigen (both primary and secondary), reagents, stabilizers and reaction buffers, tracking dies, protocols required to perform the assay, and notes on interpreting the results.
ELISA kits also contain non-reusable polystyrene microtiter plates pre-coated with a “capture” antibody, so that the reaction products are immobilized by immunosorption onto the plate. During the assay, any antibodies left unbound will be washed away. An ELISA kit typically contains enough materials to test samples in a 96-well plate.
Standard methods for quantifying the results of an ELISA are biotin, horseradish peroxidase or alkaline phosphatase (AP), all of which cause enzyme-linked color changes in the presence of the target antigen, and which can be read by spectrophotometry.
ELISA kits are used to detect food allergens, diseases, toxins, or recreational/prescription drugs, using organic samples such as urine and blood. Separate ELISA tests can be multiplexed onto a single biochip to make testing for a wider range of substances more efficient.
Other available kits include PCR amplification, functional assays and Chemiluminescent Immunoassay (CLIA) kits, which are similar to ELISA kits, but use a chemiluminescent substrate to detect or measure the amount of a target molecule.
